Okay, to put it shortly, I've been curious about Meloetta EX's Round attack. It's says "Does 30 damage times the number of your Pokemon that have the Round attack." Do the Pokemon have to be in play? The card move doesn't say otherwise.

Yes, they do. Pokemon in the discard pile never have their attacks or Abilities counted unless the card specifically mentions it (Exeggcute PLF's Propagation, Ho-Oh-EX DRX's Rebirth, Night March).
