is my subject today. I am very confused on the damage counter/poison aspect of the game.

Does damage from poison multiply with weakness? Say Mewtwo ex ndx

takes damage and poison from this Crobat. Does only the direct damage from the attack ultra-toxic fang multiply or does the poison also multiply? Yielding 80 damage and 8 damage counters? Or 80 damage and 4 damage counters?

With poison when do damage counters turn into actual damage?

Additionally if the Mewtwo doesn't retreat during his round. The poison it takes back to my turn is 1 damage counter or 4 damage counters.

and you can add 4 more counters per turn? When does the damage count. I'm very confused about the ruling that poison only dies damage counters and not actual damage. Why was that ruling needed?

As for poison counting as dealing damage, no, it does not. It clearly states on the cards that you place damage counters. Placing damage counters is not dealing damage, thereby not affected by weakness/resistance. Poison damage happens between turns, so if the opponent doesn't remove the status condition during their turn (by evolving, effect of a card like busybody ability audino, retreating, using switch/escape rope/keldeo ex's rush in ability to end up on the bench) then they receive poison damage after their turn.
As for hypnotoxic laser, that answer of weakness should be clear as it's A- answered above as to poison not being affected by weakness and B- it being the effect of a trainer card, thereby not havig a type to even be considered for weakness/resistance.
 
I believe now it is time to use MATH!

Mewtwo has weakness to Psychic, so the actual damage of Ultra-Toxic Fang is doubled - to 80. Poison is not part of an attack's damage, so it's done separately (and thus you never apply W&R), so Mewtwo has 4 damage counters put on it in between turns. However, if Mewtwo becomes Poisoned by a new source (such as Hypnotoxic Laser), apply the new source for Poison - i.e. 1 damage counter, or more only if it says otherwise. (If Virbank City Gym is in play, add 2 more damage counters to whatever it is regularly, so 4+2=6 in between turns, or 1+2=3 for HTL.)

So as long as Mewtwo remains Poisoned via the effect of Ultra-Toxic Fang, put 4 damage counters on it in between turns (6 if Virbank is around at the time). If Mewtwo moves to the Bench, it's no longer Poisoned, so it needs to be re-applied. If Mewtwo is poisoned via another source, you go by the new number.
