I think all that's important is that the Marker is properly drawn out and centered (for the most part; seems it doesn't need to be perfect perfect) on a black background. The border around the black background needs to be white. There's a video of people in Japan, who made a giant AR card, like 50 feet by 50 feet, something like that. They brought it to a big hangout/meetup/central park-ish kind of thing, and they had one of them go into a building with a 3DS, and made a giant Samus from Metroid, and it was bigger than the nearby buildings!
Apparently, Nintendo designated June 25th at Nintendo 3DS StreetPass Day and left it to everyone to select the location within their area. You pretty much meet other 3DS owners and use StreetPass. What you do is up to everyone participating. I don't think Nintendo has anything formally planned for the day, as they're not formally sponsoring and specific event.
