From the link Arceus mentioned, specifically you want to look at Tournament Rules, Tournament Formats, Operation Procedures, and Penalty Guidelines. There's also a Professor Program mission statement that you'll want to look at, found
here - you should also know the Core Values.
In addition, you'll want to be hip on your rulings, so I also recommend viewing the
Compendium. This mostly covers the rulings on how cards interact, from the simple to the complicated. Compendium LV.X is what you want to look at the most, since it's a bit more complete. Compendium BW does have a few things, but most of what's in there is already in Compendium LV.X.
The test is about 45 minutes long and there's 40 questions, which gives you an average of just over a minute per question. Try to pace yourself. Answer all the questions as you go through, and if you have time, when you're done, go over your answers again to double-check.
A passing score is 32 questions correct or more (80%). If you don't get 32, you can take the exam again after 2 weeks (or is it 1 month? it's one of them). Most people are comfortable on the rulings section, but when they get to the judging bits, that's where they tend to flub up the most, so don't skimp on studying that.
It's not a really hard test, but it is reasonably challenging. Best of luck!
